چکیده

This introduction to the R package mixlow is a (slightly) modified version of Boik and Narasimhan (2010), published in the Journal of Statistical Software. Synergistic and antagonistic drug interactions are important to consider when developing mixtures of anticancer or other types of drugs. Boik, Newman, and Boik (2008) proposed the MixLow method as an alternative to the Median-Effect method of Chou and Talalay (1984) for estimating drug interaction indices. One advantage of the MixLow method is that the nonlinear mixed-effects model used to estimate parameters of concentrationresponse curves can provide more accurate parameter estimates than the log linearization and least-squares analysis used in the Median-Effect method. This paper introduces the mixlow package in R, an implementation of the MixLow method. Results are reported for a small simulation study.

Perspectives in Pharmacology Revisiting the Isobole and Related Quantitative Methods for Assessing Drug Synergism

The isobole is well established and commonly used in the quantitative study of agonist drug combinations. This article reviews the isobole, its derivation from the concept of dose equivalence, and its usefulness in providing the predicted effect of an agonist drug combination, a topic not discussed in pharmacology textbooks. This review addresses that topic and also shows that an alternate meth...

Studies on antibiotic synergism and antagonism: the effect in vitro of combinations of antibiotics on bacteria of varying resistance to single antibiotics.

Earlier studies on combined antibiotic action from this laboratory have resulted in the formulation of a scheme for combined antibiotic action. Antibiotics have been divided tentatively into two groups: I. penicillin, streptomycin, bacitracin, and neomycin; II. chloramphenicol, aureomycin, terramycin (Jawetz and Gunnison, 1952a,b; Jawetz et al., 1952). Mixtures of group I drugs often resulted i...

Reconceptualizing synergism and antagonism among multiple stressors

The potential for complex synergistic or antagonistic interactions between multiple stressors presents one of the largest uncertainties when predicting ecological change but, despite common use of the terms in the scientific literature, a consensus on their operational definition is still lacking. The identification of synergism or antagonism is generally straightforward when stressors operate ...
